559967 2007-06-18 11:00:00 If you are wanting to take photos (or even look at) bacteria under a microscope, you really need the 100x lens with oil immersion to see any sort of detail. Without the oil, the refractive index will be incorrect.

Don't the microscope science kits you can buy for kids educational toys contain a variety of stains ready to use?
Yes realise that. The 100x objective is oil immersion and with a 10X ocular gives 1000x magnification. Sorry I gave the magnification of the objective lenses instead of total magnification. Some kids microscope sets have simple stains but not really suitable. I want to make Loffler's Methylene Blue which when ripened give a polychrome stain forming a violet compound with polychrome qualities.
I am also interested in Algae, Protozoa and diatom specimens.

Sounds like you have it covered. I take it you have a nice stagnant pond lined up for your wildlife subjects.

Try 500mls of ethanol plus pepermint, a touch of garlic and this might get you unboggled, unboggled and reboggled.

559972 2007-06-19 12:01:00 Thanks all. Very informative. I am making Methylene Blue stain to be able to see those little nasties. Anybody out there interested in microscopy ?

You can get methylene blue at some aquarium / pet stores. It's used because it's tozic to many disease microbes but relatively harmless to Nitrobacter & Nitrosomonas which are required for aquarium filtration. Be careful with this stuff. A single drop of 5% solution in a cup of water can stain the bathroom sink blue for weeks! I know as I did it several times. The first was an accident, while the second was to see how strong it had to be to stain :D

P.S. If you see an aquariast with blue seals on their tanks, now you know why!

559973 2007-06-19 13:24:00 I have a recipe for a microscope stain which requires an item 95% ethanol. Any chemistry students out there who can tell me what it is and where to get it.

well i know of a particular drink that you can get about that strong

smells like licorice and tastes like burning:lol:
